Function

Acts as a scaffolding protein that can activate the inflammatory transcription factor NF-kappa-B and p38/JNK MAP kinase signaling pathways. Forms a signaling complex with BCL10 and MALT1, and activates MALT1 proteolytic activity and inflammatory gene expression. MALT1 is indispensable for CARD14-induced activation of NF-kappa-B and p38/JNK MAP kinases (PubMed:11278692, PubMed:21302310, PubMed:27071417, PubMed:27113748). May play a role in signaling mediated by TRAF2, TRAF3 and TRAF6 and protects cells against apoptosis. Isoform 3. Not able to activate the inflammatory transcription factor NF-kappa-B and may function as a dominant negative regulator (PubMed:21302310, PubMed:26358359).

Activity summary

CARD14 also known as CARMA2 is a protein involved in the regulation of immune responses. Its molecular weight is approximately 120 kDa. This protein is expressed mainly in keratinocytes which are cells found in the epidermis of the skin. Mechanically CARD14 functions as a scaffold protein that facilitates the assembly of signalosomes. These structures are essential for transmitting important signals within cells that activate various pathways linked to inflammation and immune responses.

Biological function summary

CARD14 plays a significant role in the NF-kB signaling pathway activating it to promote inflammation and immune responses. CARD14 forms a complex with other proteins to facilitate the activation of this pathway which leads to the expression of genes involved in inflammation and immune responses. Through this complex formation CARD14 helps modulate both the innate and adaptive immune systems by influencing the behavior of immune cells making it critical for proper skin and immune function.

Pathways

CARD14 acts as a mediator within the NF-kB and MAPK pathways both vital for cellular responses to stress cytokines and pathogens. CARD14 interacts with other proteins such as BCL10 and MALT1 which are part of the CARD-CC/BCL10/MALT1 signalosome. This interaction helps transduce signals that promote the expression of pro-inflammatory cytokines and chemokines which are critical for mounting effective immune reactions.

Associated diseases and disorders

CARD14 has strong connections to psoriasis vulgaris and pityriasis rubra pilaris. Mutations in the CARD14 gene are associated with these inflammatory skin conditions which are marked by excessive immune activation and chronic inflammation. CARD14 influences these diseases by affecting the expression of cytokines mediated through the NF-kB pathway. Its interaction with BCL10 further emphasizes its role in promoting inflammation contributing to the pathogenesis of these disorders.
