    Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Cardiac Troponin T antibody [EPR3695] (ab91605)

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human cardiac muscle tissue sections labelling Cardiac Troponin T with purified ab91605 at 1/500 dilution (0.32 μg/ml). Heat mediated antigen retrieval was performed using Heat mediated antigen retrieval using Bond™ Epitope Retrieval Solution 2 (pH 9.0) . Tissue was counterstained with Hematoxylin. Rabbit specific IHC polymer detection kit HRP/DAB (Rabbit specific IHC polymer detection kit HRP/DAB ab209101) secondary antibody was used at 1/0 dilution. PBS instead of the primary antibody was used as the negative control. The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

    Western blot - Anti-Cardiac Troponin T antibody [EPR3695] (ab91605)

The molecular weight observed is consistent with what has been described in the literature (PMID: 15793653).

    All lanes: Western blot - Anti-Cardiac Troponin T antibody [EPR3695] (ab91605) at 1/1000 dilution

Lane 1: Human heart lysate at 20 µg

Lane 2: Human skeletal muscle lysate at 20 µg

Lane 3: 293T (Human embryonic kidney epithelial cell) whole cell lysate at 20 µg

    Secondary

All lanes: Goat Anti-Rabbit IgG (HRP) with minimal cross-reactivity with human IgG at 1/2000 dilution

    Predicted band size: 35 kDa

    Observed band size: 39 kDa

    Immunoprecipitation - Anti-Cardiac Troponin T antibody [EPR3695] (ab91605)

Purified ab91605 at 1/50 dilution (2μg) immunoprecipitating Cardiac Troponin T in Human heart lysate.
Lane 1 (input): Human heart lysate 10μg
    Lane 2 (+): ab91605 + Human heart lysate.
Lane 3 (-): Rabbit monoclonal IgG (Rabbit IgG, monoclonal [EPR25A] - Isotype Control ab172730) instead of ab91605 in Human heart lysate.
VeriBlot for IP Detection Reagent (HRP) (VeriBlot for IP Detection Reagent (HRP) ab131366) (1/1000 dilution) was used for Western blotting.
Blocking Buffer and concentration: 5% NFDM/TBST.
