Supplier Data

Western blot - Anti-Carma 1/CARD11 antibody [EPR29724-541] (AB324567)

Blocking and diluting buffer and concentration : 5% NFDM/TBST.

Low expression : Neuro-2a.

Negative control : brain (PMID : 11278692).

In lanes 3-7, the lysates were stored at -80℃ prior to Western Blotting. The bands beneath the target band (150 kDa) are likey to be degradation products. In lanes 1-2, To minimize protein degradation, cells/tissues were lysed immediately after harvest and then applied to a gel and transfer membrane for Western blotting as soon as possible.

All lanes:

Western blot - Anti-Carma 1/CARD11 antibody [EPR29724-541] (ab324567) at 1/1000 dilution

Lane 1:

A20 (mouse reticulum sarcoma B lymphocyte) fresh whole cell lysate at 80 µg

Lane 2:

Neuro-2a (mouse neuroblastoma neuroblast) fresh whole cell lysate at 80 µg

Lane 3:

A20 (mouse reticulum sarcoma B lymphocyte) frozen whole cell lysate at 80 µg

Lane 4:

Neuro-2a (mouse neuroblastoma neuroblast) frozen whole cell lysate at 80 µg

Lane 5:

Mouse spleen frozen tissue lysate at 80 µg

Lane 6:

Mouse thymus frozen tissue lysate at 80 µg

Lane 7:

Mouse brain frozen tissue lysate at 80 µg

Target data

Adapter protein that plays a key role in adaptive immune response by transducing the activation of NF-kappa-B downstream of T-cell receptor (TCR) and B-cell receptor (BCR) engagement (PubMed : 12154356, PubMed : 12356734, PubMed : 16356855). Transduces signals downstream TCR or BCR activation via the formation of a multiprotein complex together with BCL10 and MALT1 that induces NF-kappa-B and MAP kinase p38 (MAPK11, MAPK12, MAPK13 and/or MAPK14) pathways (PubMed : 12154356, PubMed : 12356734, PubMed : 16356855). Upon activation in response to TCR or BCR triggering, CARD11 homooligomerizes to form a nucleating helical template that recruits BCL10 via CARD-CARD interaction, thereby promoting polymerization of BCL10 and subsequent recruitment of MALT1 : this leads to I-kappa-B kinase (IKK) phosphorylation and degradation, and release of NF-kappa-B proteins for nuclear translocation (By similarity). Its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner (By similarity). Promotes linear ubiquitination of BCL10 by promoting the targeting of BCL10 to RNF31/HOIP (By similarity). Stimulates the phosphorylation of BCL10 (By similarity). Also activates the TORC1 signaling pathway (By similarity).
