ab317147 detects CASC5 protein at kinetochore by immunofluorescent analysis. Sample : HeLa cells were fixed in 4% paraformaldehyde at RT for 15 min. Green : CASC5 stained by ab317147 diluted at 1 : 500. Red : alpha Tubulin a cytoskeleton marker stained by alpha Tubulin antibody [GT114] diluted at 1 : 1000. Blue : Fluoroshield with DAPI.

A431 whole cell and nuclear extracts (30 ug) were separated by 5% SDS-PAGE and the membrane was blotted with ab317147 diluted at 1 : 1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody and the signal was developed with Trident ECL plus-Enhanced.

CASC5 also known as KNL1 or Blinkin is a large protein with a molecular mass of approximately 250 kDa. It is an essential component of the mitotic kinetochore complex which plays an important role in chromosome segregation during cell division. CASC5 is predominantly expressed in dividing cells and localizes to kinetochores during mitosis. The protein is critical for kinetochore-microtubule attachment ensuring the correct alignment and separation of chromosomes.
Biological function summary

CASC5/KNL1 is involved in maintaining genomic stability by facilitating proper chromosome segregation. It serves as a scaffold protein and forms part of the KMN network a core microtubule-binding complex at the kinetochore. This network also includes other key proteins such as NDC80 and Mis12 complex which coordinate the attachment of spindle microtubules to chromosomes. The presence of CASC5 in this complex is essential for its stability and function impacting the proper progression of the cell cycle.

Pathways

CASC5 participates in regulatory mechanisms associated with the cell cycle and mitotic checkpoints. It integrates into the spindle assembly checkpoint pathway important for preventing aneuploidy by ensuring that cells do not undergo mitosis until all chromosomes are correctly attached to the spindle apparatus. Within these pathways CASC5 interacts with proteins such as BUB1 and MAD1 which are checkpoint regulators that monitor spindle assembly and activate the mitotic checkpoint when errors are detected.

Improper function or expression of CASC5 has been linked to various cancers. Abnormal CASC5 activity can lead to chromosomal instability which is a hallmark of cancer progression and is often observed in aggressive tumors. CASC5 also connects to the protein Aurora B kinase in cancerous cells as Aurora B is involved in correcting kinetochore-microtubule attachment errors. Additionally CASC5 mutations have been associated with microcephaly a neurodevelopmental disorder where the protein's malfunction affects the proper division and proliferation of neural progenitor cells.

Kinetochores, consisting of a centromere-associated inner segment and a microtubule-contacting outer segment, play a crucial role in chromosome segregation by mediating the physical connection between centromeric DNA and spindle microtubules (PubMed : 18045986, PubMed : 19893618, PubMed : 27881301).
