Function

Casein kinases are operationally defined by their preferential utilization of acidic proteins such as caseins as substrates (PubMed:11955436, PubMed:1409656, PubMed:18305108, PubMed:23902688). It can phosphorylate a large number of proteins (PubMed:11955436, PubMed:1409656, PubMed:18305108, PubMed:23902688). Participates in Wnt signaling (PubMed:11955436). Phosphorylates CTNNB1 at 'Ser-45' (PubMed:11955436). May phosphorylate PER1 and PER2 (By similarity). May play a role in segregating chromosomes during mitosis (PubMed:1409656). May play a role in keratin cytoskeleton disassembly and thereby, it may regulate epithelial cell migration (PubMed:23902688). Acts as a positive regulator of mTORC1 and mTORC2 signaling in response to nutrients by mediating phosphorylation of DEPTOR inhibitor (PubMed:22017875, PubMed:22017877). Acts as an inhibitor of NLRP3 inflammasome assembly by mediating phosphorylation of NLRP3 (By similarity).

Notes

Recombinant multiclonals are a mixture of recombinant antibodies co-expressed from a library of heavy and light chains.

Recombinant multiclonal antibodies offer the sensitivity of polyclonal antibodies by recognising multiple epitopes, along with consistency of a recombinant antibody.

    Immunocytochemistry/ Immunofluorescence - Anti-Casein Kinase 1 alpha Antibody [RP23040231] (ab313493)

Immunofluorescent analysis of HCT 116 cells fixed with 4% formaldehyde reconstituted in 1X PBS for 10 min at room temperature and permabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling endogenous CSNK1A1 with ab313493 at 1/100 dilution followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor 488 conjugate at 1/2000 dilution. Nuclei (blue) were stained using DAPI, and Rhodamine Phalloidin at 1/300 dilution was used for cytoskeletal F-actin (red) staining. Detection and localization of CSNK1A1 (green) in the nucleus and cytoplasm can be clearly observed in interphase cells (a-d). When cell cycle progresses to prophase (e-h) CSNK1A1 can be visualized detaching from chromatin and further progression to metaphase (i-l) and anphase and telophase (m-t) results in complete detachment of CSNK1A1 from chromatin and translocation to cytoplasm. The images were captured at 60X magnification.
