Involved in the activation cascade of caspases responsible for apoptosis execution. Might function by either activating some proteins required for cell death or inactivating proteins necessary for cell survival (PubMed:15073321). Associates with PIDD1 and CRADD to form the PIDDosome, a complex that activates CASP2 and triggers apoptosis in response to genotoxic stress (PubMed:15073321).

Lane 1: Wild-type HAP1 cell lysate (20 μg)
Lane 2: Caspase-2 knockout HAP1 cell lysate (20 μg)
Lanes 1 and 2: Merged signal (red and green). Green - ab17952 observed at 51 kDa. Red - loading control, Anti-beta Actin antibody [mAbcam 8226] - Loading Control ab8226, observed at 42 kDa.
ab179520 was shown to recognize Caspase-2 when Caspase-2 knockout samples were used, along with additional cross-reactive bands. Wild-type and Caspase-2 knockout samples were subjected to SDS-PAGE. ab179520 and Anti-beta Actin antibody [mAbcam 8226] - Loading Control ab8226 (loading control to beta Actin) were both diluted 1/1000 and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776) secondary antibodies at 1/10000 dilution for 1 h at room temperature before imaging.
