Immunocytochemistry/ Immunofluorescence - Anti-Caspase 8 (phospho S387) antibody [RP23040092] (AB308013)
For immunofluorescence analysis, HeLa cells with TNF-alpha at 20 ng/mL for 20 minutes were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.25% Triton X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature for detection of Caspase 8 (phospho S387) using ab308013 at 1/500 dilution in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor® 488 Goat anti-Rabbit IgG Secondary Antibody at a dilution of 1/400 for 30 minutes at room temperature (Panel a : green). F-actin (Panel b : red) was stained with Alexa Fluor® 594 Phalloidin Nuclei (Panel c : blue) were stained with DAPI Panel d is a merged image showing nuclear localization respectively and e shows competition with the Caspase 8 (phospho S387) peptide. The images were captured at 20X magnification.

Western blot - Anti-Caspase 8 (phospho S387) antibody [RP23040092] (AB308013)
Western blot analysis of Caspase 8 (phospho S387) was performed by loading 20 µg of Jurkat tissue lysates untreated and treated with TNF-alpha (20 ng/mL for 30 minutes) lysates (lane 1, 2) using a 4-12% Bis-Tris gel, electrophoresis system, and pre-Stained Protein Standard. Proteins were transferred to a nitrocellulose membrane and blocked with 5% skim milk for 1 hour at room temperature. Caspase 8 (phospho S387) was detected at ~55 kDa using ab308013 at 1/1000 dilution in 2.5% skim milk at 4°C overnight on a rocking platform. To confirm specificity, competition was performed with the phosphopeptide (10 µg/mL) (lane 3, 4). Goat anti-Rabbit IgG - HRP Secondary Antibody at 1 : 5000 dilution was used and chemiluminescent detection was performed (ECL).
