CATSPER3 also known as Cation Channel Sperm-Associated Protein 3 functions primarily as a calcium selective ion channel. This protein with a molecular weight of approximately 85 kDa forms part of the CATSPER channel complex located on the flagellar membrane of sperm cells. Expression of CATSPER3 occurs mainly in the testis. The channel plays a critical role in mediating calcium influx necessary for sperm motility and fertilization capabilities.
Biological function summary

CATSPER3 contributes significantly to sperm function. It forms a complex with other CATSPER proteins — CATSPER1 CATSPER2 and CATSPER4 — that combine to create a functional ion channel. This channel regulates calcium entry in response to triggers such as progesterone important for hyperactivation of sperm cells an enhanced motility state that facilitates penetration through the egg's zona pellucida. Disruption in any part of this complex impairs sperm function and can affect fertility.

CATSPER3 operates within the pathway controlling sperm motility often influenced by the CatSper signaling pathway. This pathway governs calcium ion entry a pivotal event for initiating and sustaining sperm hyperactivated motility. CATSPER3 interacts closely with accessory proteins including CATSPERβ and CATSPERG supporting proper localization and functioning of the CATSPER ion channel within the sperm tail.

CATSPER3 has associations with male infertility particularly in cases related to asthenozoospermia a condition marked by reduced sperm motility.
