Flow Cytometry (Intracellular) - Anti-CBL (phospho Y700) antibody [EP2225Y] - BSA and Azide free (AB284732)
This data was developed using the same antibody clone in a different buffer formulation (ab76002).
Flow cytometry analysis of 4% paraformaldehyde fixed 90% methanol permeabilized Jurkat cells (human T cell leukemia T lymphocyte from peripheral blood) labelling CBL with ab76002 at 1/1000 dilution, followed by blocking with 10% goat serum for 1h at room temperature. ab76002 was incubated for 30 min at room temperature (RT) and then Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150081) was incubated for 30 min at room temperature. Positive staining on Jurkat cells treated with 1mM pervanadate for 30min imatinib (middle) while negative with treated with 1mM pervanadate for 30min followed by Alkaline phosphatases (right) treatment.

Untreated Jurkat (Human T cell leukemia T lymphocyte) whole cell lysate at 15 µg
Lane 2:
Jurkat treated with 1mM pervanadate for 30min whole cell lysate at 15 µg
Lane 3:
Jurkat treated with 1mM pervanadate for 30min whole cell lysate, then the membrane was incubated with alkaline phosphatase at 15 µg

Biological function summary
The CBL protein acts as a negative regulator for several signaling pathways. It integrates into signaling complexes and modulates the activation of pathways by interacting with other proteins. CBL does not act alone; it commonly forms complexes with proteins such as Grb2 and p85 influencing downstream signaling influence in pathways. This regulation prevents excessive activation of signals that can lead to cellular proliferation or differentiation abnormally.
Pathways
CBL integrates into the ubiquitin-proteasome system and the Ras signaling pathway. Within these pathways CBL works to modulate cell surface receptors ensuring that signaling threshold is not surpassed.
