CBLN1 also known as C1q-like protein 4 is a protein of interest in the nervous system. It has a mass of approximately 33 kDa and is expressed largely in the cerebellum and other regions of the brain. CBLN1 plays a role in synapse formation and maintenance. It serves as a ligand to neurexins and other synaptic receptors facilitating synaptic integrity and plasticity essential for normal brain function.
Biological function summary

The function of CBLN1 is associated with the maintenance of neuronal communication. It is an integral part of a complex that includes the presynaptic neurexins and postsynaptic gridhub proteins contributing to synaptic organization. This protein ensures proper connectivity and signaling at the synaptic junctions which is critical for cognitive and motor activities.

Pathways

CBLN1 is involved in essential synaptic pathways critical for communication between neurons. It plays a significant part in the formation and maintenance of excitatory synapses working closely with neurexin proteins. CBLN1 is also associated with pathways involving cerebellar development where it interacts with cerebellar granule cells and their synaptic partners guiding proper synaptic targeting and function.

CBLN1 connections have implications in neurodegenerative diseases like cerebellar ataxias and schizophrenia. Disruption in the function or expression of CBLN1 can lead to synaptic dysfunction contributing to the pathology of these disorders. Among associated proteins dysfunctional interactions with GRID2 or neurexins have been observed in relation to these diseases linking the synaptic deficits observed with CBLN1 abnormalities.

Required for synapse integrity and synaptic plasticity. During cerebellar synapse formation, essential for the matching and maintenance of pre- and post-synaptic elements at parallel fiber-Purkinje cell synapses, the establishment of the proper pattern of climbing fiber-Purkinje cell innervation, and induction of long-term depression at parallel fiber-Purkinje cell synapses. Plays a role as a synaptic organizer that acts bidirectionally on both pre- and post-synaptic components. On the one hand induces accumulation of synaptic vesicles in the pre-synaptic part by binding with NRXN1 and in other hand induces clustering of GRID2 and its associated proteins at the post-synaptic site through association of GRID2. NRXN1-CBLN1-GRID2 complex directly induces parallel fiber protrusions that encapsulate spines of Purkinje cells leading to accumulation of GRID2 and synaptic vesicles. Required for CBLN3 export from the endoplasmic reticulum and secretion (By similarity). NRXN1-CBLN1-GRID2 complex mediates the D-Serine-dependent long term depression signals and AMPA receptor endocytosis (PubMed : 27418511). Essential for long-term maintenance but not establishment of excitatory synapses (By similarity). Inhibits the formation and function of inhibitory GABAergic synapses in cerebellar Purkinje cells (By similarity).. The cerebellin peptide exerts neuromodulatory functions. Directly stimulates norepinephrine release via the adenylate cyclase/PKA-dependent signaling pathway; and indirectly enhances adrenocortical secretion in vivo, through a paracrine mechanism involving medullary catecholamine release (By similarity).
