CBX7 also known as Chromobox protein homolog 7 is a member of the Polycomb group proteins. It has a molecular mass of approximately 27 kDa. Mechanically CBX7 functions as a transcriptional repressor that interacts with chromatin to maintain the repression of genes involved in development and cell differentiation. The protein is expressed in various tissues with notable levels in embryonic stem cells and certain adult tissues like bone marrow and the immune system.
Biological function summary

The CBX7 protein participates in the complex known as Polycomb Repressive Complex 1 (PRC1). This complex plays a significant role in the maintenance of cellular identity by regulating gene expression profiles important for stem cell pluripotency and differentiation. CBX7 binds to modified histones leading to chromatin compaction and gene silencing instrumental for preserving cell lineage integrity during development and cellular transformation.

Pathways

CBX7 interacts with several components in the cell cycle regulation and epigenetic modification pathways. Within the cell cycle regulation pathway it closely interacts with other Polycomb group proteins to control gene expression that governs cell proliferation and differentiation. In the context of epigenetic pathways CBX7 influences chromatin remodeling by interacting with proteins like BMI1 and RING1 thereby exerting control over the transcriptional landscape required for developmental and adaptive responses.

CBX7 shows a relationship with cancer and developmental disorders. Aberrant expression or mutation of CBX7 has been associated with several cancers including prostate and gastric cancers often connected with the suppression of tumor suppressor genes and cell cycle regulation. In these diseases CBX7 interacts with proteins like E2F6 and MYC which are important for cellular proliferation and growth. Moreover disruptions in CBX7 expression patterns can lead to improper cell differentiation contributing to developmental abnormalities.

Component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. Promotes histone H3 trimethylation at 'Lys-9' (H3K9me3). Binds to trimethylated lysine residues in histones, and possibly also other proteins.
