CCK4 also known as PTK7 protein is a receptor tyrosine kinase. The molecular mass of the CCK4 protein is approximately 118 kDa. It is expressed in various tissues including the placenta lung kidney and colon. The presence of CCK4 is particularly noted in developmental tissues and has roles in embryogenesis. Despite its classification as a receptor tyrosine kinase CCK4 lacks catalytic activity indicating its unique functional properties.
CCK4 participates in cellular signaling involved in adhesion and cell migration. It localizes predominantly in the membranes participating in complex cellular environments such as the Wnt signaling complex. This suggests a role in maintaining cellular architecture and signaling pathways that influence developmental processes. Its interactions within these complexes can help maintain balance and functional integrity in developing tissues.
CCK4 engages in vital pathways influencing cell adhesion and movement including the planar cell polarity (PCP) pathway and Wnt signaling pathway. Within these pathways CCK4 interacts with proteins such as Dishevelled and Frizzled establishing its role in guiding cellular and developmental patterns. These interactions regulate important aspects of cell polarity and directional movement during embryonic development.
CCK4 has been implicated in cancer and congenital malformations. In cancers such as colorectal carcinoma altered CCK4 expression correlates with cancer progression possibly through interaction with associated proteins like ROR2. CCK4 was immunoprecipitated from Jurkat (human T cell leukemia cell line from peripheral blood) whole cell lysate (prepared using NETN buffer; 1 mg for IP, 20% of IP loaded) with ab225874 at 6 μg/mg lysate. Western blot was performed from the immunoprecipitate using ab225874 at 1 μg/ml.
