Immunocytochemistry/ Immunofluorescence - Anti-CCL3 / MIP-1 alpha antibody [EPR19900-147] - BSA and Azide free (AB251590)

Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized THP-1 (human monocytic leukemia cell line) cells, treated with 80nM TPA overnight, then treated with 100ng/ml LPS for 3h and together with 300ng/ml BFA for another 3h or Untreated control, labeling Macrophage Inflammatory Protein 1 alpha / CCL3 with ab245251 at 1/100 dilution followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution (green). Confocal image showing cytoplasmic staining on THP-1 cells treated with 80nM TPA overnight, then treated with 100ng/ml LPS for 3h and together with 300ng/ml BFA for another 3h.
The nuclear counter stain is DAPI (blue). Tubulin is detected with Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) (ab195889) (red) at 1/200 dilution.
Secondary antibody only control : Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution.

Intracellular flow cytometric analysis of 4% paraformaldehyde-fixed, 0.1% Tween-20-permeabilized THP-1 (human monocytic leukemia cell line) cell line, treated with 80nM TPA overnight, then treated with 100ng/ml LPS for 3h and together with 300ng/ml BFA for another 3h (red) or Untreated control (green), labeling Macrophage Inflammatory Protein 1 alpha / CCL3 with ab245251 at 1/500 dilution (red) compared with a Rabbit IgG, monoclonal [EPR25A] - Isotype Control (ab172730) (black) and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) at 1/2000 dilution was used as the secondary antibody.

Biological function summary
Proteins including MIP-1 alpha participate in the recruitment and activation of diverse immune cells aiding the inflammatory response. Through interaction with receptors like CCR1 and CCR5 MIP-1 alpha mobilizes cells like natural killer cells and monocytes. This chemokine does not generally form part of a larger protein complex but interacts directly with membrane receptors to exert its function in immune cell chemotaxis.
