Chemotactic for lymphocytes and monocytes. Is a ligand for CCR1, CCR3 and CCR5. Is an inhibitor of HIV-1-infection. The processed form LD78-beta(3-70) shows a 20-fold to 30-fold higher chemotactic activity and is a very potent inhibitor of HIV-1-infection. LD78-beta(3-70) is also a ligand for CCR1, CCR3 and CCR5.

Activity summary

CCL3L1 also known as chemokine (C-C motif) ligand 3-like 1 is a protein encoded by the CCL3L1 gene. This chemokine weighs approximately 10 kDa. CCL3L1 binds to chemokine receptors CCR1 and CCR5 on the surface of target cells. It is secreted by various cells such as macrophages T-cells and dendritic cells. Mainly it expresses in tissues involved in immune responses including lymph nodes and spleen.

Biological function summary

CCL3L1 plays an important role in mediating immune responses. It attracts immune cells like monocytes neutrophils and T-lymphocytes to sites of infection or injury enhancing the immune defense. Although CCL3L1 does not form part of a larger protein complex it acts as a critical signaling molecule in the immune system. Its interaction with CCR5 makes it an important component in inflammatory responses and leukocyte chemotaxis.

Pathways

CCL3L1 functions as a significant participant in cytokine-cytokine receptor interactions and chemokine signaling pathways. These pathways regulate the movement and activity of immune cells in response to specific chemokine signals. Through these pathways CCL3L1 interacts with other chemokines such as CCL4 supporting the coordination of immune cell trafficking to inflamed tissues. This involvement is essential for maintaining the proper function of the immune response.

Associated diseases and disorders

CCL3L1 is associated with susceptibility to HIV infection and rheumatoid arthritis. Variations in the CCL3L1 gene copy number can alter individual risk to HIV as this protein competes for binding with the HIV-1 virus on the CCR5 receptor effectively impeding viral entry. Additionally the overexpression of CCL3L1 contributes to the inflammatory environment observed in rheumatoid arthritis by recruiting more immune cells to affected joints and it interacts with proteins like TNF-α in the inflammatory cascade.
