CCM2 also known as Malcavernin functions in the stability of cell-cell junctions in blood vessels. It has an approximate molecular mass of 50 kDa. The protein is largely expressed in vascular endothelial cells which are cells lining the inside of the blood vessels. CCM2 forms part of a protein scaffold that guides the cellular responses to mechanical stress maintaining vascular integrity.
Biological function summary

It interacts with several proteins to form a well-coordinated complex including KRIT1 and CCM3 contributing to the maintenance of vascular structure. The CCM2 protein is essential for regulating the cellular pathways that respond to oxidative stress. It facilitates actin cytoskeleton organization and modulates endothelial barrier function providing a structural basis for cellular processes involving vascular homeostasis.

Pathways

The protein is an important component in the RhoA signaling and MAPK pathways which influence cellular shape and proliferation. Within these pathways CCM2 influences various proteins including ROCK and MEK to execute its function. It ensures proper signal transduction that is necessary for maintaining cellular responses to environmental changes which can be important for normal vascular function.

CCM2 mutations associate closely with cerebral cavernous malformations (CCM) which trigger abnormal blood vessel formation in the brain that leads to seizures or stroke. During this condition the protein's interaction with KRIT1 exhibits significant influence since defects in the gene encoding CCM2 or KRIT1 can lead to disease phenotypes. Researchers study these interactions to better understand and develop therapeutics for vascular malformation-related diseases.
