High affinity receptor for the C-C type chemokines CCL17/TARC, CCL22/MDC and CKLF isoform 1/CKLF1. The activity of this receptor is mediated by G(i) proteins which activate a phosphatidylinositol-calcium second messenger system. Can function as a chemoattractant homing receptor on circulating memory lymphocytes and as a coreceptor for some primary HIV-2 isolates. In the CNS, could mediate hippocampal-neuron survival.

CCR4 also known as C-C chemokine receptor type 4 is a G protein-coupled receptor with the molecular mass of approximately 41 kDa. It is predominantly expressed on the surface of T-helper 2 cells regulatory T cells and certain dendritic cells. CCR4 functions by binding chemokines such as CCL17 and CCL22 facilitating chemotaxis of immune cells to sites of inflammation. Researchers often refer to antibodies targeting CCR4 like mogamulizumab when studying immune responses.
CCR4 plays an important role in the trafficking of immune cells to inflammatory sites therefore regulating immune response and tolerance. It is part of the chemokine receptor family which is integral to the immune system's function. CCR4 helps maintain the balance between effector and regulatory T cells contributing to immunological homeostasis. Its involvement in migration and positioning of immune cells within lymphoid and peripheral tissues highlights its importance in maintaining immune surveillance.
CCR4 significantly influences the chemokine signaling pathway which is pivotal in directing the movement and positioning of immune cells. This receptor interacts with key proteins like CCL17 and CCL22 to mediate cell chemotaxis in these pathways. Additionally CCR4 is associated with the T-cell receptor signaling pathway where it coordinates with other receptors and proteins to modulate immune responses.
Increased CCR4 expression has connections with various conditions such as adult T-cell leukemia/lymphoma and cutaneous T-cell lymphomas. In these diseases CCR4 assists tumor cells in evading the immune system fueling disease progression. Proteins like TARC (thymus and activation-regulated chemokine) and MDC (macrophage-derived chemokine) ligands for CCR4 often facilitate tumor cell migration and proliferation. Therapeutic approaches targeting CCR4 such as the anti-CCR4 antibody mogamulizumab have shown potential in treating these malignancies by disrupting these interactions.

10% SDS PAGE gel
A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CCR4 is 37.7kDa.
All lanes: Western blot - Anti-CCR4 antibody [KW-0761 (Mogamulizumab)] - Rabbit IgG (Chimeric) (ab275980) at 2 µg/mL
Lane 1: Human cerebellum tissue lysate at 35 µg
Lane 2: Human heart tissue lysate at 35 µg
Predicted band size: 41 kDa
False colour image of Western blot: Anti-CCR4 antibody [KW-0761 (Mogamulizumab)] - Chimeric staining at 2 ug/ml, shown in green; Mouse anti-Alpha Tubulin [DM1A] (Anti-alpha Tubulin antibody [DM1A] - Loading Control ab7291) loading control staining at 1/20000 dilution, shown in red. In Western blot, ab275980 was shown to bind specifically to CCR4. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween$®$ 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L 800CW and Goat anti-Mouse IgG H&L 680RD at 1/20000 dilution.
All lanes: Western blot - Anti-CCR4 antibody [KW-0761 (Mogamulizumab)] - Rabbit IgG (Chimeric) (ab275980) at 2 µg/mL
