Supplementary information

CCT6A also known as TCP-1-zeta or CCT-zeta acts as a component of the chaperonin-containing TCP-1 complex with a mass approximately 60 kDa. This complex facilitates protein folding in an ATP-dependent manner. Predominantly researchers find CCT6A in the cytoplasm of eukaryotic cells and it expresses in various tissues indicating its broad functional role.
Biological function summary

CCT6A plays an important role in folding newly synthesized proteins and refolding denatured proteins. It is a part of the essential chaperonin complex that consists of eight different subunits including CCT6A which collaborates to ensure correct protein conformation. This function supports cellular proteostasis maintaining the balance and health of the proteome which is critical for proper cell function and survival.

Pathways

CCT6A participates in the protein folding pathway and interacts with the protein catabolic processes. It plays an important part in pathways such as cytosolic protein folding and folding of actin and tubulin. Through these pathways CCT6A associates with proteins like HSP70 which also contributes to protein folding and stability signifying shared roles in maintaining cell homeostasis.

CCT6A's involvement impacts various neurodegenerative conditions and cancer. Misfolded proteins that are not adequately managed by chaperonin complexes including CCT6A are linked to diseases like Alzheimer's. In cancer the misregulation of protein folding pathways involving CCT6A might contribute to tumorigenesis. CCT6A holds connections with proteins such as p53 where impaired folding of tumor suppressors can lead to oncogenic outcomes.

Target data

Component of the chaperonin-containing T-complex (TRiC), a molecular chaperone complex that assists the folding of proteins upon ATP hydrolysis (PubMed : 25467444). The TRiC complex mediates the folding of WRAP53/TCAB1, thereby regulating telomere maintenance (PubMed : 25467444). The TRiC complex plays a role in the folding of actin and tubulin (Probable).
