Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human tonsil tissue labelling CD103/Integrin alpha E with ab129202 at a dilution of 1 : 1000. Heat mediated antigen retrieval was performed using AR9 antigen retrieval solution and microwave treatment for 15 min at 20% power. Anti-Rabbit/Mouse HRP polymer (PerkinElmer Opal Polymer HRP Ms Plus Rb) was used as secondary antibody. Opal tyramide amplification was performed using Opal 520 fluorophore. Counterstained with DAPI stain. Image scanned with Vectra 3.0 and analyzed via Phenochart software.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human colon tissue labeling CD103 with purified ab129202 at 1/500. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9. ab97051 an HRP-conjugated goat anti-rabbit IgG (H+L) was used as the secondary antibody (1/500).
Negative control using PBS instead of primary antibody (inset).
Counterstained with hematoxylin.

Immunohistochemical analysis of formalin fixed paraffin embedded human colon labelling CD103 with ab129202 at a concentration of 0.5µg/ml. The immunostaining was performed on a Ventana DISCOVERY ULTRA (Roche Tissue Diagnostics) instrument with a OptiView DAB IHC Detection Kit. Heat mediated antigen retrieval was performed with DISCOVERY cell conditioning solution (CC1) 100°C, pH8.5 for 32mins.
ab129202 anti-CD103 antibody [EPR4166(2)] was incubated for 16mins at 37°C. Sections were counterstained with Hematoxylin II. Image inset shows absence of staining in secondary antibody only control.
Customers are encouraged to optimise antigen retrieval conditions, antibody concentration, incubation times and temperature for best results in their own IHC assay workflow (automated and manual)

Immunohistochemistry analysis (Formalin/PFA-fixed paraffin-embedded sections) of formaldehyde-fixed human Tonsil tissue. Stained with ab129202 at 1/1000 dilution. Secondary antibody used was Rabbit IgG VisUCyte HRP Polymer Antibody. Blocking was done with Bloxall Endogenous Peroxidase and Alkaline Phosphatase Blocking for 10 minutes at 25°C. The sample was incubated with the primary antibody and 1% BSA in PBS Buffer for 24 hours at 4°C. Antigen retrieval method was heat mediated, Citrate pH6.

Biological function summary
CD103 is essential for establishing and maintaining interactions between immune cells and epithelial cells. By incorporating into the αEΒ7 integrin complex it plays an important role in the localization of intraepithelial lymphocytes and dendritic cells in the gut mucosa and skin contributing to immune surveillance and protection of these barriers. CD103 helps to control lymphocyte activation and the migration process offering a critical surface marker for identifying tissue-resident memory T cells (Trm cells) and discerning their functional potential.
Pathways
CD103 functions prominently in the immunological environment by participating in pathways that regulate lymphocyte adherence and migration. Two significant pathways it fits into include the T-cell homing and retention pathways and the antigen presentation pathway. Through these pathways CD103 interacts with molecules such as E-cadherin and impacts the movement of T cells to epithelial sites influencing tissue-specific immune responses alongside related proteins like integrin beta 7 which forms a part of the same integrin complex.
