Flow Cytometry - Anti-CD200 / OX2 antibody [MRC OX90] - BSA and Azide free (AB244564)
This data was developed using the same antibody clone in a different buffer formulation (ab33734)
C57BL/6 mouse splenocytes stained with ab33734 (right) or Rat IgG2aκ (ab18450) isotype (left). C57BL/6 mouse splenocytes were incubated for 30 min on ice in 1x PBS / 10 % mouse serum to block FC receptors and non-specific protein-protein interaction followed by the antibody (ab33734) or Rat IgG2aκ (ab18450) (1x106 in 100 μl at 0.2 μg/ml) for 30 min on ice.
The secondary antibody Goat anti-rat IgG H&L (Alexa Fluor ® 488, pre-adsorbed) (ab150165) was used at 1/2000 dilution for 30 min on ice. The cells were simultaneously stained with CD19 antibody.
Acquisition of >30,000 events were collected using a 50 mW Blue laser (488nm) and 530/30 bandpass filter. Events were gated on viable lymphocytes.

Spleen cells from immunised rats were fused with cells of the rat Y3 myeloma cell line.

Biological function summary
CD200 regulates the immune system by inhibiting inflammatory responses and maintaining immune homeostasis. It does not act as part of a significant multi-protein complex but directly interacts with CD200R. This interaction promotes an anti-inflammatory state and is important in preventing autoimmune diseases and excessive tissue damage. The expression of CD200 is also an indicator of immune tolerance in various tissues.
Pathways
CD200 is involved in immune regulatory pathways. It plays a pivotal role in the suppression of immune responses through the CD200-CD200R signaling pathway. This pathway is vital for dampening inflammatory processes related to autoimmune reactions and is consequently linked to the maintenance of tissue homeostasis. Proteins related to CD200 through these pathways include CD200R and other molecules involved in immune checkpoint pathways.
