Immunocytochemistry - Anti-CD204 antibody [004] - BSA and Azide free (AB277135)
Confocal immunofluorescence analysis of CD204 in RAW264.7 (mouse macrophage cell line transformed with Abelson murine leukemia virus cells. Cells were fixed with 4% PFA, blocked with 10% serum, and incubated with ab277135 at 1/100 dilution at 4°C overnight. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue).

Flow Cytometry - Anti-CD204 antibody [004] - BSA and Azide free (AB277135)
Flow cytometric analysis of CD204 expression on Raw264.7 (mouse macrophage cell line transformed with Abelson murine leukemia virus) cells. Cells were stained with ab277135 at 1/25 dilution, then a FITC-conjugated second step antibody (Black). Cells were also staibed with an isotype control (Grey). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.
- IHC-P

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-CD204 antibody [004] - BSA and Azide free (AB277135)
Formalin-fixed, paraffin-embedded embedded mouse liver tissue stained for CD204 using ab277135 at 1/100 dilution in immunohistochemical analysis.

Biological function summary
CD204 influences inflammatory responses and lipid metabolism. It does not function alone but as part of a complex involving several other molecules like pattern recognition receptors. CD204 recognizes and binds to modified lipoproteins and is significant in processes like phagocytosis and clearance of cellular debris. Its activity affects immune response modulation and can influence atherosclerosis development by its interaction with oxidized low-density lipoproteins.
Pathways
The role of CD204 extends to key cellular and immunological pathways. This receptor is involved in the Toll-like receptor signaling pathway which is important for innate immunity besides participating in the macrophage activation pathways. In these pathways CD204 interacts with other proteins including CD36 and SRA. This interaction helps coordinate inflammatory responses and the resolution of inflammation.
