Function

Could play a role in phagocytic activities of tissue macrophages, both in intracellular lysosomal metabolism and extracellular cell-cell and cell-pathogen interactions. Binds to tissue- and organ-specific lectins or selectins, allowing homing of macrophage subsets to particular sites. Rapid recirculation of CD68 from endosomes and lysosomes to the plasma membrane may allow macrophages to crawl over selectin-bearing substrates or other cells.

Activity summary

CD68 also known as KP1 or ED1 is a glycoprotein mainly expressed on the surface of macrophages and monocytes. Structurally it is a member of the lysosome-associated membrane protein (LAMP) family and has a molecular weight of approximately 110 kDa. This protein is often used as a marker in immunohistochemistry (IHC) and immunofluorescence staining to identify macrophages in various tissues. Researchers rely on anti-CD68 antibodies for Western blot and other analytical techniques to study its expression and function.

Biological function summary

CD68 participates in the innate immune response by allowing macrophages to bind internalize and process various antigens. This protein does not function as part of a larger complex but plays a role in processes such as phagocytosis and cytokine release. Its involvement in antigen presentation suggests a role in both initiating and regulating immune cell responses. Through these mechanisms CD68 supports the clearance of pathogens and debris from the body’s tissues.

Pathways

Researchers have highlighted that CD68 plays a significant part in the inflammatory response and atherogenesis. In these pathways CD68 interacts with C1q and other scavenger receptors on macrophages to mediate lipid uptake and foam cell formation. The protein is also connected to the regulation of cytokines like IL-1 and IL-6 which are critical in inflammatory signaling pathways promoting further immune responses.

Associated diseases and disorders

Studies indicate that CD68 expression is prominent in atherosclerosis and certain types of cancer such as lymphoma. In atherosclerosis CD68-expressing macrophages accumulate lipids leading to foam cell formation and plaque development. Moreover in cancerous conditions CD68 often correlates with tumor-associated macrophages (TAMs) playing a role in tumor progression and immune evasion. In these disorders CD68 frequently interacts with proteins involved in chronic inflammation offering potential targets for therapeutic interventions.

    ab955 staining CD68 in human liver tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).

    Tissue was fixed with paraformaldehyde. Samples were then blocked with 10% serum for 3 hours at 22°C followed by incubation with the primary antibody at a 1/100 dilution for 16 hours at 4°C. An Alexa-Fluor® 568 conjugated goat anti-mouse polyclonal was used as secondary antibody at a 1/400 dilution.

    ab955 staining CD68 in human ulcerated oral (Mucosa/Bone) tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections).

    Tissue was fixed with paraformaldehyde, permeabilized with 0.1% Triton-X 100 in PBS and blocked with 2.5% serum for 90 minutes at 25°C; antigen retrieval was by heat mediation in citrate buffer (pH 6). Samples were incubated with primary antibody (1/500 in 1% serum in PBS +0.01% Triton-X 100) for 16 hours at 4°C. A commercial IHC kit and DAB was used to visualize the staining.

    Anti-CD68 antibody [RM1031] ab303565 works better than ab955 in western blot testing.

    We suggest optimizing experimental protocols (increasing lysate amount, using lower dilution or higher sensitivity ECL substrate) to improve results when using ab955 in western blot.

    Immunohistochemical analysis of formalin fixed paraffin embedded human liver labelling CD68 with ab955 at a concentration of 0.1 µg/ml. The immunostaining was performed on a Ventana DISCOVERY ULTRA (Roche Tissue Diagnostics) instrument with a OptiView DAB IHC Detection Kit. Heat mediated antigen retrieval was performed with DISCOVERY cell conditioning solution (CC1) 100°C, pH8.5 for 32mins.

    ab955 Anti-CD68 antibody [KP1] was incubated for 16mins at 37°C. Sections were counterstained with Hematoxylin II. Image inset shows absence of staining in secondary antibody only control.

    Customers are encouraged to optimise antigen retrieval conditions, antibody concentration, incubation times and temperature for best results in their own IHC assay workflow (automated and manual)
