Immunofluorescence analysis of CD73 was performed using 70% confluent log phase U-87 MG cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with ab54217 at a concentration of 1 μg/mL in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with a Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 conjugate, (1 : 2000 dilution), for 45 minutes at room temperature (Panel a : Green). Nuclei (Panel b : Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI F-actin (Panel c : Red) was stained with Rhodamine Phalloidin (1 : 300 dilution). Panel d represents the merged image showing membrane localization. Panel e represents HeLa cells showing no expression of CD73. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Biological function summary
CD73 participates in the regulation of purinergic signaling through adenosine production. It acts in cooperation with other cell surface enzymes such as CD39 forming a functional complex that handles ATP breakdown to adenosine. This enzymatic activity modulates immune responses tissue protection and inflammation control. In addition to immune-related functions CD73 supports the maintenance of vascular integrity and proper cellular adhesion which is important in different physiological and pathological contexts.
Pathways
CD73 plays an important part in the adenosinergic pathway governing the conversion of AMP into adenosine. This activity impacts the signaling and function of the adenosine receptors on immune and non-immune cells. Another important pathway is the angiogenesis process where CD73 contributes by affecting endothelial cell migration and blood vessel development. Relationships with key proteins such as adenosine A2 receptors help facilitate these critical pathway activities.
