Activity summary

CD75 also known as ST6GAL1 is an enzyme that belongs to the glycosyltransferase family specifically classified as a sialyltransferase. CD75 catalyzes the addition of sialic acid to glycoproteins and glycolipids impacting their function and stability. It has an approximate molecular mass of 45 kDa. CD75 is expressed in a variety of cell types including B lymphocytes and epithelial cells. Its expression can be observed on the cell surface where it plays important roles in cell-cell interactions.

Biological function summary

CD75 contributes to the mechanics of cell signaling and immune system modulation. CD75 is involved in the biosynthesis of sialylated glycoconjugates which affect cellular recognition and communication. This enzyme does not form part of a multi-protein complex but interacts with other proteins upon glycosylation events. Through its action CD75 impacts multiple cellular processes including inflammation regulation and pathogen recognition.

Pathways

CD75 plays roles in glycosylation and immune response mechanisms. It participates in the N-glycan biosynthesis pathway which influences protein folding and stability. Additionally CD75 impacts the sialylation pathway where it modifies the surface proteins for lymphocyte activation. Importantly CD75 interacts with proteins such as Siglecs which are involved in immune response modulation and cellular recognition.

Associated diseases and disorders

CD75 is implicated in cancer progression and autoimmune diseases. In various cancers such as colon cancer altered CD75 expression affects metastatic potential and tumor cell survival. Additionally aberrant CD75 expression occurs in autoimmune diseases like lupus wherein it modulates immune cell activity. CD75 interacts with proteins like CD22 a sialic acid-binding lectin on B cells influencing disease pathogenesis and progression through immune system pathways.

    ab27316, at a dilution of 10µl for 106 cells or 100 µl of whole blood, staining CD75 by Flow Cytometry.

    CD75 was immunoprecipitated using 0.5mg HepG2 whole cell extract, 5μg of Mouse monoclonal to CD75 and 50μl of protein G magnetic beads (+). No antibody was added to the control (-).

    The antibody was incubated under agitation with Protein G beads for 10min, HepG2 whole cell extract lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.

    Proteins were eluted by addition of 40μl SDS loading buffer and incubated for 10min at 70°C; 10μl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab27316.
