Flow Cytometry - Anti-CD80 antibody [EPR22127-251] - BSA and Azide free (AB238649)
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab222702).
Flow cytometry overlay histogram showing mouse splenocytes treated with 10µg/m LPS for 3 Days (bottom) and negative untreated mouse splenocytes (top) stained with ab222702 within the CD11c+ sub-population. The cells were incubated in 1x PBS containing 10μg/ml anti CD16/CD32 and 10% normal goat serum to block FC receptors and non-specific protein-protein interactions, followed by the antibody (ab222702) (1x 106 in 100μl at 0.04 μg/ml (1/12,500 dilution)) for 30min on ice.
The secondary antibody Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed was incubated at 1/4000 for 30min on ice.
Isotype control antibody (black line) was Recombinant Rabbit IgG, monoclonal [EPR25A] - Isotype Control used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.
Acquisition of >5000 events were collected using a 50 mW Blue laser (488nm) and 525/40 bandpass filter.

Flow cytometric analysis of P815 (mouse mastocytoma mast cell line, left) cells and C2C12 ( mouse myoblast, right) cells labeling CD80 with ab222702 at 1/5000 (red) compared with Rabbit IgG, monoclonal [EPR25A] - Isotype Control (ab172730) (black) and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077), at 1/2000 dilution was used as the secondary antibody.
Gated on viable cells.

Biological function summary
CD80 plays a significant role in the immune system's ability to distinguish between self and non-self. It belongs to the B7 family and participates in the formation of immunological synapses between T cells and antigen-presenting cells. This interaction is essential for triggering T cell proliferation and cytokine production forming a part of the adaptive immune response. CD80's expression increases in response to pathogen recognition enhancing the immune system's efficiency.

CD80 is integral to the regulation of T cell-mediated immune responses. It is involved in the CD28 co-stimulatory pathway which is vital for T cell activation and function.
