Function

Plays a role in a late step of leukocyte extravasation helping cells to overcome the endothelial basement membrane. Acts at the same site as, but independently of, PECAM1 (By similarity). Homophilic adhesion molecule, but these interactions may not be required for cell aggregation (By similarity).

Activity summary

CD99L2 also known as CD99-like 2 is a transmembrane protein with an approximate mass of 32-35 kDa. It shares structural similarities with the well-characterized CD99 protein. CD99L2 can be found across various tissue cell types including immune cells and endothelial cells. The protein's mechanical role involves aiding in the transendothelial migration of leukocytes which is an essential step in immune surveillance and response.

Biological function summary

CD99L2 participates in the dynamic regulation of leukocyte extravasation. This protein is not just a single actor but part of a larger set of molecules that facilitate the movement of white blood cells from the bloodstream to sites of infection or injury. CD99L2 interacts with other proteins to form complexes that enable the effective rolling adhesion and transmigration of leukocytes across the endothelial barrier.

Pathways

CD99L2 integrates functionally within the leukocyte adhesion and diapedesis pathways. It works closely with other key molecules such as ICAM-1 and VCAM-1 mediating efficient leukocyte passage through endothelial layers. In these pathways CD99L2 cooperates with signaling proteins and other adhesion molecules to maintain proper immune responses forming a network critical for cellular traffics such as inflammatory responses.

Associated diseases and disorders

CD99L2 displays a connection to inflammatory diseases and certain immune system dysfunctions. Changes or disruptions in CD99L2 expression or function can be related to chronic inflammatory conditions like rheumatoid arthritis where excessive or inappropriate immune cell infiltration can occur. CD99L2's interaction with related adhesion molecules such as its homologue CD99 indicates its role in epidermal permeability barrier function abnormalities showing relevance to conditions like psoriasis.
