Cdc23 also known as ANAPC8 is an essential component of the anaphase-promoting complex/cyclosome (APC/C). This protein has a molecular mass of approximately 64 kDa and plays an important role in controlling the cell cycle by functioning as an E3 ubiquitin ligase. Cdc23 tags target proteins for degradation through the ubiquitin-proteasome system ensuring proper cell cycle progression especially during the transition from metaphase to anaphase. It is widely expressed in proliferating cells reflecting its significant role in cell division processes.
Biological function summary

Cdc23 contributes to the regulation of cell cycle progression. It is a part of the larger APC/C complex which oversees the degradation of cell cycle regulators such as securin and cyclins. The function of Cdc23 is key in the timely progression from metaphase to anaphase guaranteeing that chromosomes segregate accurately. This regulation protects genomic integrity and is necessary for normal cell proliferation and growth.

Pathways

Cdc23 participates actively in the regulation of the cell cycle and the ubiquitin-proteasome pathway. It works closely with other components of the APC/C complex to modulate the activity of cyclin-dependent kinases (CDKs). This modulation ensures proper turnover of critical proteins coordinating cell cycle events. Cdc20 and Cdh1 are two other regulatory proteins that interact with Cdc23 within these pathways providing temporal control to the cell division process.

Cdc23 is connected to cancer and some neurodegenerative disorders. Aberrations in its function or expression levels can lead to uncontrolled cell growth commonly associated with different cancers. The misregulation of the APC/C complex involving Cdc23 can also impact neuronal cells linking its role to neurodegenerative diseases like Alzheimer's. The interactions with other APC/C complex proteins such as Cdc27 further illustrate the critical roles of Cdc23 in maintaining normal cellular and physiological functions.

Component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle (PubMed : 18485873). The APC/C complex acts by mediating ubiquitination and subsequent degradation of target proteins : it mainly mediates the formation of 'Lys-11'-linked polyubiquitin chains and, to a lower extent, the formation of 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains (PubMed : 18485873). The APC/C complex catalyzes assembly of branched 'Lys-11'-/'Lys-48'-linked branched ubiquitin chains on target proteins (PubMed : 29033132).
