Cdc27 also known as cell division cycle 27 or APC3 plays an important role in cell cycle regulation. It weighs approximately 92 kDa and is expressed in various tissues although it is found predominantly within the nucleus of proliferating cells. Cdc27 is a component of the anaphase-promoting complex/cyclosome (APC/C) which is vital for progression through mitosis and the G1 phase of the cell cycle by targeting specific proteins for degradation.
Biological function summary

Cdc27 serves as an essential part of the APC/C a large E3 ubiquitin ligase complex. By forming the scaffold for other subunits Cdc27 helps regulate the ubiquitination and subsequent degradation of mitotic cyclins and other proteins involved in cell cycle progression. The interaction of Cdc27 with other proteins like Cdc16 and Cdc23 stabilizes the APC/C complex ensuring its proper function.

Pathways

Cdc27 participates actively in the cell cycle especially during the G2/M transition and mitotic exit. It is involved in the ubiquitin-proteasome pathway where it intersects with proteins like securin and cyclin B1. These interactions allow Cdc27 to mediate the transition between cell cycle phases and support chromosome segregation.

Disruptions in Cdc27 expression or function are linked to cancer development particularly breast and colorectal cancers. Abnormal activity in the APC/C complex can lead to uncontrolled cell proliferation. Component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle (PubMed : 18485873). The APC/C complex acts by mediating ubiquitination and subsequent degradation of target proteins : it mainly mediates the formation of 'Lys-11'-linked polyubiquitin chains and, to a lower extent, the formation of 'Lys-48'- and 'Lys-63'-linked polyubiquitin chains (PubMed : 18485873).
