Immunocytochemistry/ Immunofluorescence - Anti-Cdk9 antibody (AB38840)
ICC/IF image of ab38840 stained human HeLa cells. The cells were PFA fixed (10 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab38840, 1μg/ml) for 1h at room temperature. 1%BSA / 10% normal serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Cdk9 antibody (AB38840)
IHC image of ab38840 staining Cdk9 in Human breast adenocarcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab38840, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Biological function summary
Cdk9 functions as a central component of transcriptional processes. It is essential for the transition of RNA polymerase II from the initiation phase to productive elongation. As part of the P-TEFb complex Cdk9 targets negative elongation factors helping relieve their inhibition. This regulation is especially important in the control of genes with strong promoter proximally paused polymerase. By doing so Cdk9 controls the expression of several genes involved in cell growth differentiation and response to stress.
Pathways
Cdk9 plays a significant role in transcription and signal transduction pathways. One important pathway is the NF-κB signaling where Cdk9 regulates the transcription of NF-κB target genes involved in immune response. Another significant pathway is the heat shock response where Cdk9 enhances the expression of heat shock proteins helping cells deal with stress. In both pathways it cooperates with other proteins like cyclins and negative elongation factors illustrating its interconnected role in cellular function.
