Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-CDS2 antibody (AB234973)

Paraffin-embedded human liver tissue stained for CDS2 with ab234973 at 1/200 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody.

Supplementary information

CDS2 also known as CDP-diacylglycerol synthase 2 is an enzyme that plays an important role in lipid metabolism. This protein has a molecular mass of approximately 52 kDa. It catalyzes the conversion of phosphatidic acid to CDP-diacylglycerol which is an important precursor for the synthesis of many phospholipids. CDS2 is mainly expressed in tissues with high lipid turnover such as the liver brain and adipose tissues.
Biological function summary

CDS2 is involved in maintaining cellular membrane integrity and function. It participates in the synthesis of phosphatidylinositol which is a component of the membrane and contributes to cellular signaling. CDS2 does not act alone; it functions as part of larger lipid synthesis pathways although it does not form a specific complex with other proteins. Still it interacts functionally with enzymes involved in lipid biosynthesis to ensure efficient lipid production within the cell.

Pathways

CDS2 functionally interacts in the glycerophospholipid biosynthesis and phosphatidylinositol signaling pathway. These pathways are central to generating phosphatidylinositol and related molecules which are critical for cellular processes such as membrane trafficking and signal transduction. CDS2's activity connects it with other proteins like phosphatidylinositol synthase which further modifies products of CDS2's reaction in the cell.

Abnormalities in CDS2 expression or function have correlations with metabolic disorders such as non-alcoholic fatty liver disease. Altered lipid metabolism impacts liver function where CDS2 plays an important role. Dysregulation in lipid pathways involving CDS2 may also be linked to neurological disorders given its significant expression in brain tissues. Disease associations may involve proteins such as glycerol-3-phosphate acyltransferase as their activities are interconnected in maintaining lipid homeostasis.

Target data

Catalyzes the conversion of phosphatidic acid (PA) to CDP-diacylglycerol (CDP-DAG), an essential intermediate in the synthesis of phosphatidylglycerol, cardiolipin and phosphatidylinositol (PubMed : 25375833). Exhibits specificity for the nature of the acyl chains at the sn-1 and sn-2 positions in the substrate, PA and the preferred acyl chain composition is 1-stearoyl-2-arachidonoyl-sn-phosphatidic acid (PubMed : 25375833). Plays an important role in regulating the growth and maturation of lipid droplets which are storage organelles at the center of lipid and energy homeostasis (PubMed : 26946540, PubMed : 31548309).
