Supplementary information

The CELF3 protein also known as CUGBP Elav-like family member 3 belongs to the CELF family of RNA-binding proteins. With a molecular weight of approximately 55 kDa CELF3 localizes mainly in the brain and nervous tissue. It functions by binding to RNA which affects the stability and translation of the RNA molecules. This protein can also regulate alternative splicing contributing to the processing diversity of pre-mRNA molecules within cells.
Biological function summary

RNA-binding proteins such as CELF3 play significant roles in various cellular processes including gene expression regulation and maintaining RNA stability. CELF3 is not part of a larger complex but interacts with different RNA targets to influence their splicing patterns. In the nervous system CELF3 contributes to neuronal development and synaptic function impacting the fine-tuning of gene expression required for these processes.

Pathways

The RNA-binding activity of CELF3 impacts the mRNA processing pathway which is essential for mRNA maturation and function. It shows considerable overlap with other CELF family proteins like CELF1 and CELF2 that also partake in post-transcriptional regulation. CELF3’s role in alternative splicing also intertwines with the Wnt signaling pathway a pathway important for neuronal differentiation and development highlighting its regulatory importance in neurogenesis.

Aberrant expression or mutations of CELF3 correlates with neurological conditions such as epilepsy and neurodegenerative diseases. These associations manifest through altered RNA processing implicating CELF family members like CELF1 in these pathologies. CELF3’s involvement in splicing regulation further connects it to muscular dystrophy as misregulation may lead to improper muscle function seen in this disorder.

Target data

RNA-binding protein involved in the regulation of pre-mRNA alternative splicing. Mediates exon inclusion and/or exclusion in pre-mRNA that are subject to tissue-specific and developmentally regulated alternative splicing. Specifically activates exon 5 inclusion of cardiac isoforms of TNNT2 during heart remodeling at the juvenile to adult transition. Activates the splicing of MAPT/Tau exon 10. Binds to muscle-specific splicing enhancer (MSE) intronic sites flanking the alternative exon 5 of TNNT2 pre-mRNA.
