CENP-W also known as Centromere Protein W is a component of the CENP-A nucleosome-associated complex (NAC) and it plays an important role in centromere structure and function. This protein has a molecular weight of approximately 19 kDa. CENP-W is mostly expressed in the nucleus where it functions at the centromere. This protein is a part of the larger centromere complex and is essential in maintaining the stability of kinetochore microtubule attachments during cell division.
Biological function summary

CENP-W is necessary for the correct assembly of the kinetochore complex. It partners with other proteins within the NAC such as CENP-X and CENP-T to ensure that the centromere is properly structured for chromosomal segregation. CENP-W directly interacts with CENP-A to facilitate the recruitment of other centromere proteins ensuring cohesion and accurate segregation of chromosomes. This activity is critical for maintaining genomic stability during cell replication and division.

Pathways

CENP-W is significant in the cell cycle checkpoint regulation and mitotic pathways. CENP-W contributes to the proper activation of the spindle assembly checkpoint a critical quality control step that ensures chromosomes are correctly aligned before cell division proceeds. Component of the CENPA-NAC (nucleosome-associated) complex, a complex that plays a central role in assembly of kinetochore proteins, mitotic progression and chromosome segregation (By similarity). The CENPA-NAC complex recruits the CENPA-CAD (nucleosome distal) complex and may be involved in incorporation of newly synthesized CENPA into centromeres (By similarity). Part of a nucleosome-associated complex that binds specifically to histone H3-containing nucleosomes at the centromere, as opposed to nucleosomes containing CENPA. Component of the heterotetrameric CENP-T-W-S-X complex that binds and supercoils DNA, and plays an important role in kinetochore assembly. CENPW has a fundamental role in kinetochore assembly and function. It is one of the inner kinetochore proteins, with most further proteins binding downstream. Required for normal chromosome organization and normal progress through mitosis.
