CENPH also known as Centromere Protein H is part of the inner kinetochore complex which plays an important role in chromosome segregation during cell division. The protein has a molecular mass of approximately 29 kDa. CENPH is expressed primarily in dividing cells including those in the bone marrow and lymphoid tissues. High expression levels are observed in cancerous tissues suggesting a link to cell proliferation.
Biological function summary

CENPH plays an essential role in the assembly and stabilization of the kinetochore complex. It forms part of the CENP-A nucleosome-associated complex (NAC) critical for kinetochore function. CENPH ensures accurate attachment of spindle microtubules to the kinetochore which is necessary for proper chromosome alignment and segregation. The presence of CENPH in the kinetochore safeguards against chromosomal instability.

Pathways

CENPH contributes significantly to the cell cycle and mitotic spindle checkpoint pathways. It interacts intricately with other centromere proteins like CENP-C and CENP-A coordinating the dynamics of kinetochore and centromere function during cell division. Disruptions in any of these interactions can lead to errors in chromosome segregation which highlights CENPH's importance within these pathways.

CENPH has been associated with cancer and genetic instability disorders. Aberrant expression or mutation in CENPH can lead to missegregation of chromosomes a hallmark of cancerous cells. The CENPA-NAC complex recruits the CENPA-CAD (nucleosome distal) complex and may be involved in incorporation of newly synthesized CENPA into centromeres.
