Supplementary information

CEP120 also known as Centrosomal Protein 120 is a protein in humans encoded by the CEP120 gene. It has a molecular mass of about 121 kDa. CEP120 is primarily expressed in the centrosome an important organelle in cells responsible for microtubule organization. This protein is critical in the regulation of centrosome duplication and spindle assembly during cell division ensuring proper chromosome segregation.
Biological function summary

CEP120 plays a significant role in centrosome and cilia formation. It is an essential part of the core centrosome complex and directly interacts with other centrosomal proteins required for centriole elongation. Its activity ensures centrosome stability and proper cilia function impacting cellular sensory and signaling processes.

Pathways

CEP120 is implicated in microtubule-related pathways. It integrates into the centrosome duplication pathway closely interacting with proteins like CPAP (CENPJ) and STIL which are central to centrosome biogenesis. CEP120 also influences the pathway of ciliogenesis alongside proteins such as IFT88 being essential in the construction of primary cilia.

CEP120 mutations have been linked to ciliopathies such as Jeune asphyxiating thoracic dystrophy and some forms of microcephalic primordial dwarfism. Abnormal CEP120 expression or function is related to defective cilia and centrosome activities affecting processes like ciliary signaling. The protein interacts with OFD1 and alterations in this interaction may exacerbate these conditions illustrating its connection within the broader landscape of ciliary and developmental disorders.

Target data

Plays a role in the microtubule-dependent coupling of the nucleus and the centrosome. Involved in the processes that regulate centrosome-mediated interkinetic nuclear migration (INM) of neural progenitors and for proper positioning of neurons during brain development. Also implicated in the migration and selfrenewal of neural progenitors. Required for centriole duplication and maturation during mitosis and subsequent ciliogenesis (By similarity). Required for the recruitment of CEP295 to the proximal end of new-born centrioles at the centriolar microtubule wall during early S phase in a PLK4-dependent manner (PubMed : 27185865).
