Involved in the detoxification of xenobiotics and in the activation of ester and amide prodrugs. Shows low catalytic efficiency for hydrolysis of CPT-11 (7-ethyl-10-[4-(1-piperidino)-1-piperidino]-carbonyloxycamptothecin), a prodrug for camptothecin used in cancer therapeutics.

CES3 also known as carboxylesterase 3 or TG hydrolase (TGH) is an enzyme with a molecular mass of approximately 60 kDa. This protein functions by breaking down ester bonds effectively hydrolyzing lipids into free fatty acids and glycerol. CES3 is mainly expressed in the liver but you can also find it in other tissues including the intestine and adipose tissue. It can act on a broad range of substrates due to its versatile enzymatic activity.
CES3 plays an integral role in lipid metabolism. It is not typically a part of multi-protein complexes but works independently to hydrolyze triglycerides. By doing so CES3 helps release stored energy in the form of free fatty acids which are important for various metabolic processes. This enzyme regulates energy balance and lipid storage impacting lipid signaling pathways within cells.
CES3 participates actively in lipid metabolism and energy homeostasis pathways. It ties closely to the lipolysis pathway where it works along with proteins like hormone-sensitive lipase to mobilize stored fats for energy. Moreover CES3 interacts with lipid biosynthesis pathways linking lipid storage with breakdown and playing a significant role in maintaining lipid equilibrium in the body.
Alterations in CES3 function relate to conditions such as obesity and non-alcoholic fatty liver disease (NAFLD). Reduced or excessive CES3 activity might affect lipid metabolism thereby contributing to abnormal fat accumulation. CES3 is connected to hepatic lipase which also influences lipid processing in the liver demonstrating the enzyme’s potential impact on liver health and systemic lipid balance.
