Product details

ab288597 is a BSA and Azide Free antibody supplied in an unconjugated format and it is suitable for sandwich ELISAs to quantify Mouse CGRP. The recommended pair for sandwich ELISA is:
Capture: ab288597, Mouse CGRP Capture Antibody (unconjugated)
Detector: ab288615, Mouse CGRP Detector Antibody (unconjugated)
The reference range value is 3.91-500 pg/mL.

Sandwich ELISA
The recommended antibody orientation is based on internal optimization for ELISA-based assays. Antibody orientation is assay dependent and needs to be optimized for each assay type. Please note that the range provided for this antibody is only an estimation based on the performance of the product using the recommended antibody pair. Performance of the antibody pair will depend on the specific characteristics of your assay. We guarantee the product works in sandwich ELISA, but we do not guarantee the sensitivity or dynamic range of the antibody in your assay.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Supplementary information

Calcitonin Gene-Related Peptide I (CGRP-I) also known as CGRP-alpha is a neuropeptide with a mass of around 3.8 kDa. It is mainly expressed in the central and peripheral nervous systems particularly in sensory nerves and the trigeminal ganglia. The peptide operates mechanically as a potent vasodilator which means it can widen blood vessels by relaxing the smooth muscle cells within the vessel walls. CGRP-I achieves this by binding to receptors on the cell surface initiating a cascade of intracellular events that lead to vessel relaxation.
Biological function summary

This neuropeptide plays a significant role in the modulation of pain transmission and blood flow regulation. CGRP-I does this by promoting vasodilation and influencing synaptic transmission. It is not part of a larger complex but functions effectively through receptor interaction. In neurons it can enhance excitability and amplify pain signals which contributes to its role in the sensory system. The peptide works in a fine balance with other neurotransmitters to maintain homeostasis in the nervous system.

Pathways

CGRP-I is integral to the nociception and vasodilation pathways which are key in the body's response to pain and blood pressure modulation. In the nociception pathway CGRP-I operates alongside substances like substance P which together can potentiate pain perception. In the vasodilation pathway CGRP-I works with nitric oxide to regulate vascular tone and maintain adequate blood flow. These interactions highlight the peptide's importance in both pain modulation and cardiovascular homeostasis.

CGRP-I is closely linked to migraine and hypertension. Its relationship with migraine involves the dilation of cerebral blood vessels and facilitation of pain transmission in trigeminal nerves making it an important target for anti-CGRP antibodies in migraine therapy. CGRP-I is also associated with hypertension due to its effect on vascular tone; altered regulation can contribute to increased blood pressure. Its connection with these conditions highlights advancements in therapeutic approaches using CGRP monoclonal antibodies to manage migraines effectively and possibly benefit hypertension treatment.

Target data

CGRP1/CALCA is a peptide hormone that induces vasodilation mediated by the CALCRL-RAMP1 receptor complex (PubMed : 1318039, PubMed : 33602864, PubMed : 9620797). Dilates a variety of vessels including the coronary, cerebral and systemic vasculature. Its abundance in the CNS also points toward a neurotransmitter or neuromodulator role (PubMed : 3492492). It also elevates platelet cAMP (PubMed : 1318039). CGRP1 can also bind and activate CALCR-RAMP1 (AMYR1) receptor complex (PubMed : 38603770).
