CHAC1 also known as cation transport regulator-like protein (CHAC) plays a significant role in various cellular processes. It is a protein with an approximate mass of 27 kDa and is widely expressed in human tissues including the pancreas kidney and liver. This protein functions primarily as a γ-glutamyl cyclotransferase responsible for the cleavage of glutathione into 5-oxo-proline and cysteinylglycine therefore influencing cellular redox status and stress responses.
Biological function summary

CHAC1 acts as an important player in cell death regulation and stress response mechanisms. It participates in the unfolded protein response (UPR) during endoplasmic reticulum (ER) stress. CHAC1 engages in these processes by interacting with and being regulated by ATF4 a transcription factor activated during cellular stress. It does not function as part of a large complex but has interactions with specific stress-related proteins.

Pathways

CHAC1 is involved in the integrated stress response (ISR) pathway particularly under conditions that elicit ER stress. It has a significant role in the amino acid deprivation response pathway. The ISR pathway links CHAC1 with other proteins such as ATF4 and eIF2α and through these interactions it helps regulate cellular adaptation to stress.

CHAC1 has connections to cancer and neurodegenerative diseases. In certain cancer types CHAC1 gets upregulated linking it to the modulation of oxidative stress and apoptosis which are important processes in tumor development and progression. Additionally in neurodegenerative diseases its role in oxidative stress management potentially contributes to neuronal cell death associating it indirectly with proteins like caspase-3 that are involved in apoptotic pathways.

Catalyzes the cleavage of glutathione into 5-oxo-L-proline and a Cys-Gly dipeptide. Acts specifically on glutathione, but not on other gamma-glutamyl peptides (PubMed : 27913623). Glutathione depletion is an important factor for apoptosis initiation and execution. Acts as a pro-apoptotic component of the unfolded protein response pathway by mediating the pro-apoptotic effects of the ATF4-ATF3-DDIT3/CHOP cascade (PubMed : 19109178). Negative regulator of Notch signaling pathway involved in embryonic neurogenesis : acts by inhibiting Notch cleavage by furin, maintaining Notch in an immature inactive form, thereby promoting neurogenesis in embryos (PubMed : 22445366).
