Supplementary information

CHCHD1 also known as Coiled-Coil-Helix-Coiled-Coil-Helix Domain Containing 1 is a protein with a molecular mass of approximately 13 kDa. It is localized mainly in the mitochondria where it plays an important role in maintaining mitochondrial function. Expression of CHCHD1 is found in various tissues with an increased presence in the heart skeletal muscle and brain which indicates its functional relevance in these energy-demanding tissues.
Biological function summary

The coiled-coil structure of CHCHD1 participates in protein-protein interactions within the cell. It is a component of a larger protein complex in the mitochondrial intermembrane space which assists in maintaining the stability and function of mitochondrial proteins. CHCHD1 contributes to the regulation of mitochondrial biogenesis and dynamics affecting cellular energy production and overall metabolism.

Pathways

Studies show that CHCHD1 is involved in the mitochondrial oxidative phosphorylation pathway. It assists in regulating electron transport chain efficiency influencing ATP production. CHCHD1 also interacts with cytochrome c an important player in the electron transport chain highlighting its role in energy metabolism. These interactions ensure cells produce enough energy to support vital functions.

Research connects CHCHD1 to neurodegenerative diseases like Parkinson's and amyotrophic lateral sclerosis (ALS). Altered CHCHD1 function can disrupt mitochondrial homeostasis contributing to the pathophysiology of these conditions. Interaction with proteins such as alpha-synuclein in Parkinson's or SOD1 in ALS links CHCHD1 to these disorders emphasizing its importance in disease progression and as a potential target for therapeutic intervention.
