Function

Substrate-binding subunit of the Rab geranylgeranyltransferase (GGTase) complex. Binds unprenylated Rab proteins and presents the substrate peptide to the catalytic component B composed of RABGGTA and RABGGTB, and remains bound to it after the geranylgeranyl transfer reaction. The component A is thought to be regenerated by transferring its prenylated Rab back to the donor membrane. Besides, a pre-formed complex consisting of CHM and the Rab GGTase dimer (RGGT or component B) can bind to and prenylate Rab proteins; this alternative pathway is proposed to be the predominant pathway for Rab protein geranylgeranylation.

Activity summary

The CHM gene encodes for the Rab escort protein-1 (REP-1) which plays an essential role in the prenylation of Rab proteins a type of small GTPases involved in vesicle trafficking. The protein has a molecular mass of approximately 90 kDa. CHM is expressed in multiple tissues including the retina and nervous system. The alternate names for this protein include component of choroideremia and TCD which reflects its connection to cellular trafficking and transport processes.

Biological function summary

CHM is integral to the prenylation process which is important for anchoring Rab proteins to cellular membranes. Rab escort protein-1 acts within the Rab geranylgeranyltransferase complex facilitating the transfer of lipid groups to Rab proteins. This modification is necessary for proper Rab function affecting intracellular vesicle trafficking and membrane dynamics. CHM's biological actions are critical to cellular processes such as endocytosis exocytosis and autophagy.

Pathways

The CHM protein influences key cellular pathways such as the intracellular vesicle transport pathway and the autophagy pathway. Within these pathways CHM closely interacts with Rab proteins including Rab1 and Rab27 that are important in regulating vesicle movement within cells. The association of CHM with these pathways highlights its role in maintaining cellular transport and recycling mechanisms.

Associated diseases and disorders

CHM is strongly linked to choroideremia a genetic disorder leading to progressive vision loss. Mutations in the CHM gene disrupt the normal function of REP-1 affecting the prenylation of Rab proteins and impairing cellular processes in the retina. Additionally CHM mutations can have implications in disorders related to X-linked inheritance. Research also connects CHM with pathogenic proteins like Rab27 linked to granule release disorders highlighting the impact of its malfunction in disease development.
