Immunocytochemistry/ Immunofluorescence - Anti-Choline kinase alpha antibody (AB88053)
ICC/IF image of ab88053 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab88053, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Cells were fixed in paraformaldehyde and permeabilized with TritonX-100. A goat anti-rabbit Alexa Fluor594 was used as the secondary antibody at 1/1000 dilution.

Biological function summary
Choline kinase alpha plays a significant role in cell proliferation and survival due to its involvement in phospholipid metabolism. The enzyme is not known to be part of any large protein complexes but it does interact with a variety of cellular components that regulate its activity. It affects cellular processes by controlling the rate of phosphatidylcholine synthesis therefore impacting membrane dynamics and signal transduction. Changes in CKα activity can result in altered cell growth and function.
Pathways
Choline kinase alpha is an essential part of the Kennedy pathway or the CDP-choline pathway which is a major pathway for phosphatidylcholine biosynthesis. Within this pathway CKα's activity is essential for progressing from choline uptake to the formation of phosphatidylcholine. Other key proteins related through this pathway include CTP:phosphocholine cytidylyltransferase (CCT) and cholinephosphotransferase. The enzyme's function integrates metabolic and signaling pathways connecting lipid metabolism with cellular responses to external stimuli.
