Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Chromogranin C/SGII antibody [SGII 6B1/3] (AB20245)

IHC image of ab20245 staining in normal rat brain formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab20245, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Cells were fixed with paraformaldehyde; BSA (5%) was used as a blocking agent (40 minutes at room temperature). Secondary antibody was Goat anti-mouse IgG2b Alexa Fluor 568 Conjugation (1/600).

Chromogranin C also known as secretogranin II (SGII) is a member of the granin family and weighs approximately 68 kDa. This protein is mainly located within the dense core secretory granules of endocrine and neuronal cells. It plays a mechanical role in the storage and release of peptide hormones and neurotransmitters. Chromogranin C is found in various regions of the brain adrenal medulla and in the pituitary gland reflecting its involvement in the neuroendocrine system.
Biological function summary

This protein functions in the regulation of peptide hormone secretion and is part of a complex with other granin proteins such as Chromogranin A. It modulates the granule's environment by affecting the pH and ionic composition which influences the storage and release of hormones and neurotransmitters. Chromogranin C participates in the production of biologically active peptides after proteolytic processing playing roles in the modulation of endocrine and neuronal activities.
