Western blot - Anti-CISH/CIS antibody [RP23040117] (AB308040)

Western blot analysis was performed on whole cell extracts (30 µg lysate) of THP-1 (Lane1) and THP-1 treated with IL-2 (20 ng/24 hours) (Lane2). The blots were probed with ab308040 at 0.5-1 µg/mL and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at 1 : 2500 dilution. A clear 30 kDa band corresponding to CIS was observed in cell line tested. Known quantity of protein samples were electrophoresed using a 4-12% Bis-Tris gel, electrophoresis system and pre-stained protein standard. Resolved proteins were then transferred onto a nitrocellulose membrane. The membrane was probed with the relevant primary and secondary Antibody following blocking with 5% skimmed milk. Chemiluminescent detection was performed (ECL)

untreated THP-1 whole cell extracts at 30 µg

Lane 2:

IL-2 treated THP-1 whole cell extract at 30 µg

Target data

SOCS family proteins form part of a classical negative feedback system that regulates cytokine signal transduction. CIS is involved in the negative regulation of cytokines that signal through the JAK-STAT5 pathway such as erythropoietin, prolactin and interleukin 3 (IL3) receptor. Inhibits STAT5 trans-activation by suppressing its tyrosine phosphorylation. May be a substrate-recognition component of a SCF-like ECS (Elongin BC-CUL2/5-SOCS-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins (By similarity).
