Function

Required for checkpoint mediated cell cycle arrest in response to inhibition of DNA replication or to DNA damage induced by both ionizing and UV irradiation (PubMed:12766152, PubMed:15190204, PubMed:15707391, PubMed:16123041). Adapter protein which binds to BRCA1 and the checkpoint kinase CHEK1 and facilitates the ATR-dependent phosphorylation of both proteins (PubMed:12766152, PubMed:15096610, PubMed:15707391, PubMed:16123041). Also required to maintain normal rates of replication fork progression during unperturbed DNA replication. Binds directly to DNA, with particular affinity for branched or forked molecules and interacts with multiple protein components of the replisome such as the MCM2-7 complex and TIMELESS (PubMed:15226314, PubMed:34694004, PubMed:35585232). Important for initiation of DNA replication, recruits kinase CDC7 to phosphorylate MCM2-7 components (PubMed:27401717).

Activity summary

Claspin also known as CLSPN is a protein involved in DNA replication and checkpoint control during the cell cycle. Its estimated molecular weight is approximately 133 kDa. Claspin acts as a mediator within the ATR (ATM and Rad3-related) signaling pathway particularly during DNA damage response. The protein is highly expressed in cells with active DNA replication such as in rapidly dividing tissues.

Biological function summary

Claspin plays a significant role in stabilizing replication forks and assists in the activation of checkpoint kinase 1 (Chk1). It is not an isolated player; instead Claspin functions as part of a larger complex involving ATR and other checkpoint proteins. Through its interactions it helps to enforce a halt in the cell cycle allowing cells to repair DNA and maintain genomic integrity.

Pathways

Claspin has a critical function in the ATR-Chk1 pathway essential for the DNA damage response and the maintenance of replication fork stability. Claspin interacts with the protein TopBP1 an important player in facilitating ATR activation. It also works alongside proteins such as Rad17 and replication protein A (RPA) all of which play roles in monitoring and responding to DNA replication stress.

Associated diseases and disorders

Claspin has been linked to cancer particularly in cases where DNA damage response pathways are disrupted. Dysregulation of Claspin can affect the cell’s ability to repair DNA contributing to oncogenesis. Another condition related to Claspin is ataxia-telangiectasia a disorder characterized by total loss of ATM function which can cascade into ineffective DNA damage responses where Claspin’s role becomes compromised.
