Immunofluorescent analysis of 4% paraformaldehyde-fixed 90% confluent log phase Caco-2 cells labelling Claudin-8 with ab308007 at 1/400 dilution, followed by Alexa Fluor® 488 Goat anti-Rabbit IgG secondary antibody at a dilution of 1 : 400 (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI. Panel c is a merged image showing cell junction localization and panel d is a control without primary antibody. The images were captured using a Nikon microscope at 20X magnification.

Western blot - Anti-Claudin 8 antibody [RP23040053] (AB308007)
Western blot analysis of Claudin-8 in HeLa (lane 1) and HEK293 (lane 2) whole cell extracts using ab308007 at a dilution of 2 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~25kDa.

Western blot - Anti-Claudin 8 antibody [RP23040053] (AB308007)
Western blot analysis of Claudin-8 was performed by loading 20 µg of Jurkat, U-2 OS, HEK-293, U-87 MG, K562 and A431 cell lysates were electrophoresed using a 4-12% Bis-Tris gel, electrophoresis system and pre-stained protein standard. Resolved proteins were then transferred onto a nitrocellulose membrane. and blocked with 5% skim milk for 1 hour at room temperature. Claudin-8 was detected at ~27 kDa using ab308007 at a 1 : 1000 dilution in 2.5% skim milk at 4°C overnight on a rocking platform. Detection was performed using an HRP-conjugated Goat anti-Rabbit secondary antibody at a 1 : 5000 dilution and chemiluminescent detection (ECL).

Biological function summary
Claudin 8 contributes to the creation and maintenance of the barrier function of epithelial layers. It integrates into tight junction complexes collaborating with other claudins and occludin to establish selective paracellular transport. This organization allows for the regulation of movement of ions and small solutes between cells essential in controlling the internal environment of organs.
Pathways
Research shows Claudin 8 plays a role in epithelial and endothelial cell permeability. It is involved in the calcium signaling pathway and the MAPK (Mitogen-Activated Protein Kinase) pathway both important for cell signaling and response to extracellular stimuli. Within these pathways Claudin 8 interacts with proteins such as ZO-1 (Zonula Occludens-1) to modulate tight junction integrity and function.
