Claudin18.2 and Claudin18.1 are members of the tight junction protein family known for their role in maintaining cell-cell adhesion and sealing epithelial barriers. These proteins often referred to simply as Claudin18 have a molecular mass of approximately 26 kDa. Claudin18.2 expresses prominently in the stomach lining while Claudin18.1 is found in lung tissue. Both variants localize to tight junctions underlining their importance in forming selective cellular barriers that regulate paracellular transport.
Biological function summary

Claudin18.2 and Claudin18.1 are essential components in maintaining epithelial integrity. Claudin proteins form the backbone of tight junction strands contributing to the segregation and compartmentalization within tissues. These proteins interact with other tight junction proteins like Occludin and Zonula Occludens to form a complex which regulates permeability and protects tissues from pathogens and toxins. Their tight sealing property ensures that epithelial tissues control the passage of molecules between cells maintaining homeostasis and protecting underlying tissues from external environments.

Pathways

Claudin18.2 and Claudin18.1 integrate into the tight junction signaling pathway which is important for cell polarity and barrier function. The tight junction signaling pathway coordinates with other pathways such as the Epithelial-Mesenchymal Transition (EMT) pathway which is important in development and wound healing. Proteins like E-cadherin influence this pathway by balancing adhesion and mobility where Claudin18 proteins act to stabilize the junctions therefore controlling epithelial tissue dynamics.

Alterations in Claudin18.2 and Claudin18.1 expression associate with gastric cancer and asthma respectively. In gastric cancer overexpression of Claudin18.2 can contribute to tumor growth by facilitating cellular disorganization. It interacts with proteins like HER2 in cancer pathways suggesting a role in cell proliferation. For asthma Claudin18.1 might influence lung epithelial permeability and facilitate inflammatory processes suggesting it works with cytokines like IL-4 in the inflammatory response.
