Cleaved Caspase-7 also known as CASP-7 or cleaved Caspase is a protein involved in the execution phase of apoptosis. Its molecular weight is approximately 34 kDa before activation. It belongs to the family of cysteine-aspartic proteases and originates from a precursor with higher molecular weight. Cleavage activates it facilitating its expression in various tissues especially in locations where apoptosis is a regular process such as the liver heart and brain.
Biological function summary

Cleaved Caspase-7 plays a role by executing apoptosis through cleaving cellular substrates after aspartic acid residues. It operates independently or as part of the apoptosome complex which includes other caspases and protein factors. This complex contributes to the orderly dismantling of cellular structures during programmed cell death ensuring the removal of damaged or unnecessary cells in a controlled manner.

Pathways

Cleaved Caspase-7 participates actively in the apoptosis signaling pathway and to a lesser extent in the inflammatory response. It acts downstream in the cascade initiated by pro-apoptotic signals wherein it shares significant functional similarities with Caspase-3. Together with Caspase-8 which acts earlier in the pathway these proteins form a network that ensures the fidelity of programmed cell death. Caspase-8 initiates the cascade leading to the activation of Caspase-7.

Cleaved Caspase-7 associates notably with cancer and neurodegenerative diseases. Dysregulation of its activity often linked through aberrant apoptosis can lead to the unchecked growth of cancer cells or excessive neuronal death as seen in disorders like Alzheimer's disease. Caspase-8 is often implicated in these pathologies as well highlighting their intertwined roles in cell death and survival pathways. Consequently studying cleaved Caspase-7 helps improve understanding of and develop therapeutic strategies for these significant health issues.
