The molecular weight observed is consistent with the literature (PMID : 28459430, 30804337).

Blocking/Dilution buffer : 5% NFDM/TBST.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab222407).

The cleaved N-terminal DFNA5 also known as GSDME is a significant player in cellular processes. Upon cleaving it turns into an active fragment that carries out distinct functions. GSDME is an effector of necrotic cell death especially pyroptosis with a molecular mass of around 60 kDa. It resides within the cytoplasm of cells displaying expression in various tissues such as the colon stomach and esophagus. This protein functions as a security mechanism modulating cellular responses to stress and damage.
Biological function summary

The cleaved N-terminal fragment of GSDME disrupts the cell membrane using its pore-forming ability. GSDME belongs to the gasdermin family and becomes activated by caspase-3 a well-known executioner of apoptosis. It forms a part of a complex cellular machinery influencing the immune response and cell death. The action of GSDME in the apoptosis or pyroptosis decision is critical since it helps balance cell death pathways and inflammation preventing uncontrolled cell proliferation or excessive inflammation.

Pathways

The cleaved N-terminal GSDME integrates into the intrinsic apoptosis pathway acting downstream of caspase-3 activation. GSDME influences the balance between apoptosis and pyroptosis a critical decision point for cell fate in response to intracellular stress. It works in concert with other gasdermin family proteins such as GSDMD which plays a primary role in pyroptosis. The interaction between these proteins allows cells to select appropriate death pathways under different conditions revealing GSDME’s versatility in cellular functioning.

The function of cleaved N-terminal GSDME links to hearing loss and various cancers. Mutations in GSDME associate with autosomal dominant forms of hearing loss highlighting its role in cochlear function. Furthermore low expression levels of GSDME have connections with cancer cell survival and proliferation. This protein interacts with proteins like caspase-3 in cancer where the cleavage of GSDME may promote pyroptosis over apoptotic pathways offering insights into potential therapeutic targets for tumor management.

Gasdermin-E. Precursor of a pore-forming protein that converts non-inflammatory apoptosis to pyroptosis. This form constitutes the precursor of the pore-forming protein : upon cleavage, the released N-terminal moiety (Gasdermin-E, N-terminal) binds to membranes and forms pores, triggering pyroptosis.. Gasdermin-E, N-terminal. Pore-forming protein produced by cleavage by CASP3 or granzyme B (GZMB), which converts non-inflammatory apoptosis to pyroptosis or promotes granzyme-mediated pyroptosis, respectively (PubMed : 32188940). Mediates secondary necrosis downstream of the mitochondrial apoptotic pathway and CASP3 activation as well as in response to viral agents (PubMed : 28045099). Exhibits bactericidal activity (By similarity). Cleavage by GZMB promotes tumor suppressor activity by triggering robust anti-tumor immunity (PubMed : 32188940). Suppresses tumors by mediating granzyme-mediated pyroptosis in target cells of natural killer (NK) cells : cleavage by granzyme B (GZMB), delivered to target cells from NK-cells, triggers pyroptosis of tumor cells and tumor suppression (By similarity).
