Biological function summary
The Notch intracellular domain (NICD) which arises after further cleavage is a part of the core Notch signaling pathway. Cleaved NEAS is part of a complex involving other cleavage events; it acts as an intermediary in the Notch signaling cascade. This role makes it important for cellular communication and regulation determining cell fate decisions and controlling differentiation and proliferation. Studies often identify its presence and function within the nuclear and membrane-associated areas indicating roles both in direct gene expression and membrane signaling domains.
Pathways
Cleaved NEAS interacts heavily with Delta/Serrate/LAG-2 (DSL) Notch ligands and influences the Notch signaling pathway. The processed NICD translocates to the nucleus affecting the transcription of target genes involved in developmental processes and immune system regulation. In addition to cooperating with ligands such as Jagged and Delta-like proteins NEAS has interactions that play a part in modulating important pathways like Wnt signaling revealing its role in coordinating diverse signaling networks.
