Scavenger receptor that displays several functions associated with host defense. Promotes binding and phagocytosis of Gram-positive, Gram-negative bacteria and yeast. Mediates the recognition, internalization and degradation of oxidatively modified low density lipoprotein (oxLDL) by vascular endothelial cells. Binds to several carbohydrates including Gal-type ligands, D-galactose, L- and D-fucose, GalNAc, T and Tn antigens in a calcium-dependent manner and internalizes specifically GalNAc in nurse-like cells. Binds also to sialyl Lewis X or a trisaccharide and asialo-orosomucoid (ASOR). May also play a role in the clearance of amyloid-beta in Alzheimer disease.

Colec12 also known as Collectin-12 is a multifunctional protein involved in several biological processes. It is a member of the collectin family and often referred to as CL-P1 or SRCL. This protein has a mass of approximately 92 kDa and possesses a collagen-like region and a carbohydrate recognition domain. Expression of Colec12 occurs in various tissues including endothelial cells and macrophages where it shows specific affinity for certain types of sugars enabling it to interact with a range of microbial and host-derived molecules.
This protein contributes to innate immune responses by recognizing and binding to pathogen-associated molecular patterns (PAMPs) for pathogen clearance. Colec12 partners with other proteins as part of a larger complex facilitating the opsonization of pathogens. Through this mechanism it aids in phagocytosis whereby macrophages and other immune cells effectively clear foreign particles and dead cells. Such activity maintains tissue homeostasis and protects against infection.
Several molecular interactions underpin the role of Colec12 in innate immunity. Colec12 operates within the scavenger receptor pathway a critical part of pathogen detection making direct interactions with related proteins like mannose-binding lectin (MBL) which participate in complement activation. Furthermore Colec12 plays a role in endothelial function by binding to ligands involved in the coagulation cascade although it diverges slightly from classical coagulation pathways by engaging distinct receptors regulating immune responses.
Colec12 has been implicated in conditions such as atherosclerosis and neurodegenerative diseases. Its role in clearing oxidized low-density lipoproteins (oxLDL) links it to the development of atherosclerosis as these lipids contribute to plaque formation within arteries. Additionally how Colec12 interacts with proteins like amyloid-beta suggests a potential involvement in Alzheimer's disease. Through these associations Colec12 contributes to understanding both cardiovascular and neurodegenerative pathologies.
