This product is not recommended for use under denaturing conditions in WB. We would suggest testing it under native conditions. Denaturing and reducing conditions will greatly diminish reactivity and selectivity of this antibody. Abcam does not test ab34710 with endogenous samples in WB. We do recommend to look at the guidelines for blotting large proteins.

Anti-Collagen I + Collagen III antibody (ab34710) is stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. For extended storage, mix with an equal volume of glycerol, aliquot contents and freeze at -20° C or below.

This collagen antibody was developed using non-denatured 3D epitopes, you must be careful not to denature the collagen protein during your experiment.

It is often extremely difficult to generate antibodies with specificities to collagens due to the uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. The development of type specific antibodies is dependent on NON-DENATURED three-dimensional epitopes - this may result in diminished reactivity of some antibodies with denatured collagen or formalin-fixed, paraffin embedded tissues.

Biological function summary

Collagen I plays a central role in tissue architecture and wound healing. It is part of a larger collagen family comprising fibers that form a network within the extracellular space. In addition to its structural functions collagen I interacts with cells influencing processes like cell adhesion and migration. It undergoes remodeling through enzymes such as collagenases during tissue repair. Collagen I engages with various matrix proteins reinforcing its function in maintaining tissue homeostasis.

Activity summary

Collagen I also known as collagen type 1 is a protein that provides mechanical support to tissues. It forms large fibrils and has a molecular mass of about 300 kilodaltons. Collagen I is expressed in connective tissues including skin bones tendons and ligaments. It serves as a structural scaffold that imparts tensile strength and stability. This protein is essential for maintaining the integrity of extracellular matrices in these tissues.

Pathways

Collagen I is involved in signaling pathways related to extracellular matrix organization and remodeling. Its interactions within the transforming growth factor-beta (TGF-?) pathway highlight its role in regulating tissue repair and fibrosis. Within this pathway collagen I associates with other proteins like fibronectin. In this context matrix metalloproteinases also relate to collagen I as they modify its structure and influence matrix dynamics.

Associated diseases and disorders

Collagen I changes associate strongly with fibrosis and osteogenesis imperfecta. In fibrosis excessive collagen I accumulation leads to scarring and tissue function impairment. Proteins like TGF-? influence abnormal collagen I deposition in this context. In osteogenesis imperfecta mutations in collagen I genes disrupt its function affecting bone development and strength. Osteocalcin another bone-associated protein interacts with collagen I contributing to skeletal matrix formation.
