Immunohistochemistry (PFA perfusion fixed frozen sections) - Anti-Collagen IV antibody (AB6586)
Immunohistochemistry analysis of paraformaldehyde perfusion fixed frozen pig kidney permeabilized with 0.05% tween 20. Stained with ab6586 at 1/200 dilution. Secondary antibody used was Alexa Fluor® 647 Donkey anti-Rabbit IgG (H+L) Highly Cross-Adsorbed at 1/300 dilution. The nuclei are stained Blue with Hoechst. The Red counter stain was created with a methyl green stain and by imaging with a 555nm excitation filter. Blocking was done with Sea Block for 30 minutes at 22°C. The sample was incubated with the primary antibody and Sea Block for 16 hours at 4°C. Antigen retrieval method was heat mediated with Citrate Buffer pH 6.0.
