Function

Type IV collagen is the major structural component of glomerular basement membranes (GBM), forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen. Arresten, comprising the C-terminal NC1 domain, inhibits angiogenesis and tumor formation. The C-terminal half is found to possess the anti-angiogenic activity. Specifically inhibits endothelial cell proliferation, migration and tube formation.

Specificity

Some class-specific anti-collagens may be specific for three-dimensional epitopes which may result in diminished reactivity with denatured collagen or formalin-fixed, paraffin embedded tissues. This antibody reacts with most mammalian Type IV collagens and has negligible cross-reactivity with Type I, II, III, V or VI collagens. Non-specific cross-reaction of anti-collagen antibodies with other human serum proteins or non-collagen extracellular matrix proteins is negligible.

Purification notes

Anti-Collagen Type IV has been prepared by immunoaffinity chromatography using immobilized antigens followed by extensive cross-adsorption against other collagens, human serum proteins and non-collagen extracellular matrix proteins to remove any unwanted specificities.

Anti-Collagen IV antibody (ab6586) has 120 independent reviews from customers.

Anti-Collagen IV antibody (ab6586) specifically detects Collagen IV (UniProt ID: P02462; Molecular weight: 145kDa) and is sold in 100 µg selling sizes.

Collagen IV is a key component of the extracellular matrix in tumors, contributing to tissue stiffness and promoting cancer cell invasion. It plays a significant role in tumor progression and metastasis, influencing the behaviour of cancer cells.

Supplementary info

This supplementary information is collated from multiple sources and compiled automatically.
Activity summary

Collagen IV also known as COL4A1 and collagen type 4 plays a significant mechanical role in the structure of the basement membrane. It provides structural support and mechanical stability. Collagen IV has a molecular weight ranging approximately from 160 to 240 kDa. It is expressed in various regions including the kidney lung and vascular tissues where it contributes to the integrity and function of these organs.

Biological function summary

The primary function of collagen type IV involves forming a scaffold essential for basement membrane assembly and function. It exists as a part of a complex formation typically forming networks with other collagens and proteins. These networks are important for cellular adhesion differentiation migration and filtration processes. Collagen IV interacts with different cell surface receptors helping mediate important cellular signals.

Pathways

Collagen type 4 is integral to the extracellular matrix and integrin-mediated signaling pathways. It partners with proteins such as laminins and nidogens in these pathways contributing to cell adhesion and migration. This interaction helps regulate various cellular processes including wound healing and tissue repair. By being a part of these pathways collagen IV impacts cellular behavior and tissue homeostasis.

Associated diseases and disorders

Dysfunction or mutations in collagen IV relate to conditions such as Alport syndrome and Goodpasture's syndrome. Alport syndrome involves mutations in collagen IV leading to kidney dysfunction and hearing loss. In Goodpasture's syndrome collagen IV becomes a target for autoimmune attack resulting in damage to the lungs and kidneys. These diseases illustrate the connection between collagen IV and other proteins like laminin which together contribute to maintaining tissue integrity and preventing disease progression.
